This chapter offers a detailed overview of the structural features, different types, and chemical modifications of CDs and their derivatives, including hydroxypropyl-β-CD (HP-β-CD) and sulfobutyl ether-β-CD (SBE-β-CD). It discusses their use across various drug delivery methods such as oral, parenteral, ocular, nasal, transdermal, and rectal routes. Special focus is given to clinically approved CD-based formulations, like Captisol®-enabled products and Veklury®, which have improved therapeutic effectiveness and patient safety. The chapter also examines current regulatory views, toxicological issues, especially nephrotoxicity linked to β-CDs, and the evolving patent landscape related to CD-based biomaterials.
