Mucoadhesive hydrogel film based on cyclodextrin cross-linked with thiolated hydroxyethyl cellulose for mucosal delivery of cisplatin

The efficacy of cisplatin (CTN) is limited due to its short half-life and toxic side effects. This study aimed to synthesize mucoadhesive hydrogel film based on cyclodextrin (CD) cross-linked with thiolated hydroxyethyl cellulose (thio-HEC) for mucosal delivery of CTN.

Mucoadhesive hydrogel film based on CD cross-linked with thio-HEC was successfully generated and loaded with CTN via inclusion complex formation. Hydrogels showed 3.5-fold enhanced swelling behavior, 1623 µg/mL drug loading, and only 40 % drug released from mucoadhesive hydrogel film (H3) after 6 h. Moreover, mucoadhesive hydrogel films (H3) showed more than 75 % drug retention after 3 h on the buccal mucosa of a goat as compared to unmodified hydrogel films. Furthermore, newly designed hydrogels exhibited 11.2-fold enhanced viscosity, 7.9 N stickiness, 0.35 mJ work of adhesion, 13 mJ work of shear, and 105 mN maximum detachment force.

The newly designed mucoadhesive hydrogel film showed enhanced retention time and controlled delivery of CTN on buccal mucosa. Based on these findings, it can be concluded these newly designed mucoadhesive hydrogel film can be a vital tool for mucosal drug delivery.
