Dimethyl-α-cyclodextrin induces capacitation by removing phospholipids from the plasma membrane of mouse sperm

Time dependent effects of BSA, dimethyl alpha-CD (DMACD) and methyl beta-CD (MBCD) on the lipid efflux from sperm.. Toyoda Yokoyama Hosi medium,a modified Krebs–Ringer bicarbonate solution without BSA(fTYH) was used to incubate sperm with BSA and CDs. Cholesterol was measured by fluorescence spectrometry and phospholipids by UV/vis.

Dimethyl-alpha-CD (DMACD) was found to enhance sperm fertilization ability by removing phospholipids. It was concluded that in addition to the cholesterol acceptor albumin and methyl beta-CD (MBCD) also DMACD, not interacting with cholesterol, can induce capacitation. BSA, MBCD and DMACD increased sperm membrane fluidity, rearrangement of lipid raft and the ecrosome reaction. These findings support that phospholipid extraction is a novel trigger of capacitation.
